肠润通对糖尿病大鼠结肠糖基化终末产物及其受体表达的干预作用

Objective:The former research demonstrated that the high dosage of Changruntong(CRT) could partly restore the morphological and biomechanical reconstruction of colon in diabetic rats.In this study,the correlation between biomechanical reconstruction and expression of advanced glycation end products(AGEs) of colon in the diabetic rats was analyzed,in order to investigate the intervention mechanism of CRT. Methods:Forty SD rats were randomly divided into normal group,diabetes mellitus group,high(50g/kg) and low(25g/kg) dosage of CRT groups,and with 10 rats in each group.Rats in high and low dosage of CRT groups received gavage administration with CRT daily,and the modeling process lasted for 60 days.Blood glucose level and body weight of rats were measured termly.The expression of AGEs and RAGE in colon bowel layers was detected by using immunohistochemistry and analyzed quantificationally by using imaging analysis software.The morphological and biomechanical parameterssuch as wet weight,thickness,opening angle,residual strain and stress-strain of colonic wall were referenced to the former research reports.The correlation between the above two groups of parameters was analyzed by using linear regression analysis. Results: The expression of AGEs and RAGE in colon bowel layers of rats in DM group was higher than that of normal group(P<0.05,P<0.01),and the AGE/RAGE expressions were highly and positively correlated with morphological and biomechanical parameters.The expression of AGE and RAGE was significantly decreased in colon bowel layers of ratsin high dosage of CRT group(P<0.05,P<0.01),but the expression of AGE and RAGE in colon bowel layers of ratsin low dosage of CRT group was not changes significantly. Conclusion:The expression of AGE and RAGE in colon bowel layers is highly correlated with the biomechanical reconstruction of colonin diabetic rats,and the inhibitory effect of high dosage of CRT on AGE/RAGE expression might be one of the effective approaches for improving the morphological and biomechanical reconstruction of colon in diabetic rats.
