Bioavailability of α-tocopherol stereoisomers in sheep fed synthetic and natural α-tocopherol

Synthetic vitamin E as commercial source of supplementation of α-tocopherol has eight isomeric configurations including four 2R (RSS, RRS, RSR, RRR) and four 2S (SRR, SSR, SRS, SSS). Only the RRR stereoisomer has the highest biological activity. A ratio of 1.36:1 in relative bioavailability of RRR-α-tocopheryl acetate as a natural (Nat-α-T) source to all-rac-α-tocopheryl acetate as a synthetic (Syn-α-T) source is generally accepted, however, studies indicate either bioavailability of α-tocopherol stereoisomers or relative bioavailability between them is not constant, but rather dose dependent and differs between organs. However, no information is available about how different ratios between synthetic and natural α-tocopherol affect bioavailability. Thirty lambs were randomly assigned to five different Syn-α-T to Nat-α-T ratio including 100:0, 75:25, 50:50, 25:75 and 0:100 %. The sheep were slaughtered after 60 days of feeding and liver, heart, lungs, muscle and abdominal fat was collected for analysis for α-tocopherol stereoisomers. The data were analysed using completely randomized design. Amount of RRR-α-tocopherol generally increased in plasma and all organs, with increasing the proportion of Nat-α-T in the diet (P≤0.05). The bioavailability relative to diet of RRR-α-tocopherol in plasma, all organs and abdominal fat increased with increasing the proportion of Syn-α-T in the diet (P≤0.05). In addition, with the exception of RSR-α-tocopherol in liver, the bioavailability relative to diet of 2R-α-tocopherol in plasma, all organs and abdominal fat followed the same trend as RRR-α-tocopherol. In conclusion, the results showed that sheep express discrimination against synthetic stereoisomers of α-tocopherol in plasma and other organs.
