Project title: Water oxidation for FNM-dependent redox reactions

The oxidation of water represents an ideal reaction with respect to sustainability liberating protons and electrons and releasing oxygen. Therefore, this research project aims to apply water oxidation as the driving force for (bio)catalysis, applied in the regeneration of “consumed” Flavin mononucleotide (FMN) cofactor. Herein, light will be utilized for an in vitro biocatalytic Baeyer-Villiger oxidation reaction.

The corresponding enzymes are type II Baeyer–Villiger monooxygenases. These are two-component enzymes possessing a separated oxygenase and reductase component. This structural feature makes them highly interesting targets for the replacement of their natural reductase, being responsible for FMN recycling. Light and chemocatalysts for water oxidation will be applied in a sustainable FMN regeneration system for an enzymatic Baeyer-Villiger oxidation using in vitro lactone synthesis.

This proof-of-concept may overcome the natural dependence on nicotinamide cofactors for recycling by a direct Flavin regeneration through water oxidation. In a second step, the system will be optimized and scaled up.
