Abstract: The molecular mechanism of the sarcoplasmic reticulum Ca-ATPase has been addressed by combining information from X-ray crystallography with analysis of functional changes in site mutants. We have established a panel of assays for the partial reactions in the transport cycle of expressed wild-type and mutant Ca-ATPase, including rapid kinetic measurements of the rates of phosphorylation and dephosphorylation, conformational changes, and calcium binding and dissociation, as well as measurement of the binding affinities for nucleotides, vanadate, metal fluorides, and other inhibitors. The lecture will summarize our most recent mutational studies on the Ca-ATPase. Furthermore our assays of partial reaction steps have been adapted to analyze the mechanism of the P4-ATPase ATP8A2.
